Players can pick each other up to save them from danger or toss them into it.* Mario™, Luigi™ and two Toads are all playable characters, while many others from the Mushroom Kingdom make appearances throughout the game. Players can even ride different Yoshi characters and use their tongues to swallow enemies – or their fellow players.* In some areas, players use the motion abilities of the Wii Remote™ controller. New Super Mario Bros. Wii é uma continuação dos jogos Super Mario Bros. Assim como no original, os jogadores pulam, correm e agarram power-ups, como o Super Mushroom e a Fire Flower, que permite que eles joguem bolas de fogo em seus inimigos. Novos power-ups também foram adicionados - o Propeller Hat permite aos jogadores dar zoom verticalmente no ar e depois deslizar de volta para baixo, a Ice Flower permite que os jogadores congelem os inimigos em blocos de gelo e o Penguin Suit lhes dá tração no gelo, além de permitir para deslizar a barriga. Yoshi, de Super Mario World, também está de volta - os jogadores podem montá-lo e ele come inimigos e frutas nos níveis.
